Back when the PlayStation 2 was the world's most popular game console, the original Xbox was struggling to gain a post-Halo audience and the GameCube was a cute diversion with a couple decent titles, I trolled gaming message boards. I was so sure that anyone who bought anything other than a PS2 was out of their mind that I set on the most immature mission in history: to inform the masses, proselytize the word as handed down by Hirai-san, and ridicule those who were wasting their time with The Wind Waker.

Those who were active gamers at the time probably read that paragraph and are now either annoyed or cracking a wry smile in understanding. The latter are the ones who played Ico. Those who weren't gamers at the time can insert their own products into the mix for context.

Vision insurance and medical insurance?huh???? | Uptown Eyes ...

So with a new year, comes new insurance plans, for both?MEDICAL?as well as?VISION. ?Many do not realize these are 2 different kinds of insurance. ?I am going to try to explain what the differences are, how you can be more informed of your own plans, and how to make your exam visits flow more efficiently for you and for the clinics across the board.

Most of us have insurance benefits through our employer or we pay for our plans privately. ?Some employers offer Medical, vision, dental, and other types of insurance. ?They are typically optional plans. ?Here is the general differences of these types of insurances.

Medical Insurance plans

Medical insurance generally covers toward medical care, i.e., you go to the doctor for a problem or emergency. ?So for a general medical visit, this may consist of an ache or pain, rash, infection, or fever, just to name a few. ?Not ALL physicians are on ALL medical insurances. ?You should have been provided information regarding providers in your area, or the insurance website can be very helpful to search for one.?

Vision Insurance plans

Vision insurance generally covers?(or ?provides a benefit?)?towards eye exams and medical devices such as prescription eyewear and prescription contact lenses. ?They usually do NOT cover problem visits like, eye infections, red eyes, or other acute eye issues. ?These types of issues would be more of a medical visit and can usually be filed with your medical plan, if your eye clinic provides insurance?filing and?are providers on your plan. ?There are usually copays for most vision plans?as well as medical plans?that are due upon the time services are rendered.

Also, there are not any vision plans out?there?(that I am aware of) that will cover 100% of your vision care needs. ?Most only pay a very small percentage of your eye exam, glasses or contact lens supplies. ?Please note that most of the time, you will be responsible for overages at the time services are rendered in most eye clinics.

Dental Insurance plans

Dental plans generally covers various dental procedures. ?I am not as savvy on this particular type of plan, but I know through my personal experience, they are similar to vision plans?only cover?some things?and most expensive procedures are not billable to the dental plans OR medical plans. ?The best way to find out more about your particular dental plan is to contact your dental insurance.

Premiums, Deductibles and Co-pays

Ugh!!! ?Don?t we pay enough to these insurance companies already?! ?Most insurance plans are NOT free!! ?Someone has to pay ? the members and providers. ? Most employers offer various medical, vision, and dental benefits of some kind, at a less expensive premium (supposedly). ?You should be aware of how much is being deducted from your check monthly and know what your coverage and deductibles are. ?It may be more cost effective for you to be on your own independant insurance plan(s)?or put those premiums toward a Health Savings Account. ?These premiums that are deducted from your pay, should be pre-tax funds.

Once your coverage is activated, then you will likely owe the insurance more money before they start paying for your exams or office visits. ?This is called a deductible. ?It varies from one insurance plan to the next and they typically start over year to year. There may be an individual deductible and a family deductible. ?Either way, be aware of what your deductibles are. ?If you are not aware of them, please contact your HR department or your insurance company.

At your office visits, you may ?have a co-pay that may or may not go toward your deductible. ?This is something else you need to be aware of prior to your visits. ?Be prepared to pay your co-pays at the time the services are rendered.

How do I make life easier and my office visits more efficient?

Regardless of how you have insurance coverage, either through an employer or independantly, here are some helpful pointers to note.

- Find out the full name and contact information of the Human Resource Department and keep on file as you may need to call them a few times.
- Have ALL your insurance information available when you call to make your appointments. ?This includes subscriber/member name that is on the insurance (not nicknames), DOB (date of birth) of the member and the patient, subscriber/member ID number and patient ID number if different, Group number, insurance address and phone number.
- Be aware of your benefits. ?This can be a challenge sometimes, but you should have been provided a manual of your benefits, or have access to their website. ?If you do not have this information, it is likely that your doctor?s office does not have it either. ?Please call your HR department or call your insurance company. ?The number for members/subscribers is on the back of the card. ?Please note,?that most medical clinics do NOT have a direct line to any of the insurance companies either, so the wait time is just as long on the phone.
- Be aware if you have met your deductible or not. ?This is something your doctor?s office will definitely NOT know, so please contact your insurance company.
- Be aware of your copays or expect to pay some sort of copay. ?Most range from $10 to $50 depending on the insurance. ?Often times it will be on your card as well. ?Various clinics may be a ?specialty? clinic. ?You can call that clinic to find out what they are considered.
- Be prepared to pay 100% of the services rendered when you go to any of your doctor?s visits unless you prearranged something. ?Meaning, you called prior to your appointment and discussed this with the billing/accounting department at your doctor?s office.

Insurance myths

1. The doctor?s office gets paid by the insurance companies the difference of what I pay.

This is ABSOLUTELY FALSE. ?Most of the time, providers/physicians, take a significant?cut to be a provider listed on an insurance. ?And most of the time they don?t receive payment for 2-6 weeks after the claim is filed?sometimes longer. ?So, as you can likely imagine, the medical offices have bills to pay as well. ?In order to pay them in a timely fashion, they have to see MORE patients?(which can often times equate to less time per patient)?meaning more volume, less time/patient. ?No medical office can really afford to spend more time with patients individually since the insurance companies don?t reimburse very much, these offices have to see more patients. ?It is a sad and vicious cyle. ?This is the reason why Uptown Eyes has decided to be providers for only a few of the most common insurances plans. Our goal is to never allow insurance to dictate our time with the patient, our treatment plan, or our quality of the product we provide.??

How to OTA (over-the-air) update to iOS 6.1 on your iPhone, iPad, and iPod touch

Apple released iOS 6.1 to the public yesterday which includes the ability to buy movie tickets with Siri, better LTE support, and more. Since the introduction of iOS 5, Apple has allowed us to update our iPhones, iPads, and iPod touches over-the-air instead of having to plug into iTunes.

Normally your iPhone, iPad, or iPod touch will alert you when an update is ready, but if it hasn't, we can walk you through how to manually trigger the over-the-air update.

Note: Apple requires your battery to be at 50% or better in order to install a software update over-the-air without being plugged into a charger.

  1. Launch the Settings app from the Home screen of your iPhone, iPad, or iPod touch.
  2. Tap on General.
  3. Now tap on Software update.
  4. Your iPhone, iPad, or iPod touch will check to see if there's an update available.
  5. You'll either have the option to Download and Install or Install Now if you're device has already downloaded the software. Tap on it.
  6. You may be prompted to plug your device in to save battery. Just tap Continue.
  7. Let your iPhone, iPad, or iPod touch do its thing. It'll reboot and once it does, you should now be on the newest software version.

NEW YORK (AP) ? Amy Poehler is working on a book that appears to be a little bit of everything.

The star of "Parks and Recreation" and former "Saturday Night Live" performer has an agreement with It Books for an "illustrated, non-linear diary" with stories both true and invented.

Jokes, candor and life lessons are promised. It Books, an imprint of HarperCollins Publishers, announced Monday that the book is currently untitled and scheduled for 2014.

It's the first book for Poehler, who recently hosted the Golden Globes ceremony with Tina Fey, her good friend and author of the million-selling "Bossypants."

  • "Happy Endings"

    <strong>"Last Man Standing," ABC</strong><br /> <strong>Status: </strong> Renewed<br /> <strong>Why:</strong> Ratings for Tim Allen's return to sitcoms have been very strong, making it Tuesday's most-watched comedy. If you can beat "Glee" and "The Biggest Loser," a second season is a given.

  • "Man Up!"

    <strong>"Man Up!," ABC</strong><br /> <strong>Status:</strong> Canceled<br /> <strong>Why:</strong> ABC pulled the show after eight episodes.

  • "The Middle"

    <strong>"The Middle," ABC</strong><br /> <strong>Status:</strong> Renewed<br /> <strong>Why:</strong> Now in its third season, "The Middle" is still holding onto its middle position in the ratings on Wednesday nights, and that seems just about right. It doesn't do "American Idol" or "Survivor" numbers, of course, but with over 7 million viewers, it's a strong comedy for the network that easily beats anything NBC is offering.

  • "Missing"

    <strong>"Missing," ABC</strong><br /> <strong>Status:</strong> Canceled<br /> <strong>Why:</strong> Ashley Judd lead this drama about a former CIA agent whose son goes missing, kicking her back into action to find him. The series was only set to air 10 episodes, miniseries-style, with the potential for more, but a cancellation means that's all they're getting.

  • "Modern Family"

    <strong>"Modern Family," ABC</strong><br /> <strong>Status:</strong> Renewed<br /> <strong>Why:</strong> "Modern Family" remains ABC's biggest sitcom success story of the last decade, both in ratings and awards love. This critical darling is getting a fourth season of hijinks with the hilarious Pritchett-Dunphy clan.

  • "Once Upon A Time"

    <strong>"Once Upon A Time," ABC</strong><br /> <strong>Status:</strong> Renewed<br /> <strong>Why:</strong> "Once Upon a Time" has gotten praise for being one of the more family-friendly dramas on TV, and it's been pulling in close to 10 million viewers each week because of it. We're excited by the almost endless possibilities for new fairy tale-inspired stories to tackle in Season 2.

  • "Pan Am"

    <strong>"The River," ABC</strong><br /> <strong>Status:</strong> Canceled<br /> <strong>Why:</strong> The promise of "Paranormal Activity"-like scares each week quickly fizzled after this show premiered, along with the ratings. The first season's eight episodes came and went, and ABC has nixed any plans for more.

  • "Scandal"

    <strong>"Scandal," ABC</strong><br /> <strong>Status:</strong> Renewed<br /> <strong>Why:</strong> Shonda Rhimes' latest show isn't about doctors at all -- and that's why we're glad it's sticking around. Kerry Washington is amazing as Olivia Pope, a Washington, D.C. fixer with a crack team of specialists helping make bad headlines vanish before they're ever written. Here's hoping for more than seven episodes in its second season.

  • "Shark Tank"

  • "A Gifted Man"

    <strong>"A Gifted Man," CBS</strong><br /> <strong>Status:</strong> Canceled<br /> <strong>Why: </strong>"A Gifted Man's" total viewer numbers were OK for a Friday night, but a 1.3 rating in the 18-49 demographic is especially bad for CBS. Another bad sign: star Patrick Wilson tweeted about the end: "I had a great time. Thanks to my fans. So happy it's done." And done it is.

  • "The Amazing Race"

The show might not have the youngest audience, but it's still enough for CBS to keep it on the schedule.

  • "Criminal Minds"

  • "The Good Wife"

    Rise of Israeli centrist raises hopes for peace

    JERUSALEM (AP) ? The unexpectedly strong showing by a new centrist party in Israel's parliamentary election has raised hopes of a revival of peace talks with Palestinians that have languished for four years under Prime Minister Benjamin Netanyahu.

    Political newcomer Yair Lapid, the surprise kingmaker, is already being courted by a weakened Netanyahu, who needs his support to form a ruling coalition. Lapid has said he will not sit in the government unless the peace process is restarted.

    But following a campaign in which the Palestinian issue was largely ignored, it remains unclear how hard Lapid will push the issue in what could be weeks of coalition talks with Netanyahu.

    Tuesday's election ended in a deadlock, with Netanyahu's hard-line religious bloc of allies and the rival bloc of centrist, secular and Arab parties each with 60 seats, according to near-complete official results. Opinion polls had universally forecast a majority of seats going to the right-wing bloc.

    While Netanyahu, as head of the largest single party in parliament, is poised to remain prime minister, it appears impossible for him to cobble together a majority coalition without reaching across the aisle.

    Lapid, whose Yesh Atid ? or There is a Future ? captured 19 seats, putting it in second place, is the most likely candidate to join him. In a gesture to Netanyahu, Lapid said there would not be a "blocking majority," in which opposition parties prevent the prime minister from forming a government. The comment virtually guarantees that Netanyahu will be prime minister, with Lapid a major partner.

    Netanyahu said Wednesday he would work to create a wide coalition stretching across the political divide.

    Speaking to reporters, he said the election proved "the Israeli public wants me to continue leading the country" and put together "as broad a coalition as possible."

    He said the next government would pursue three major domestic policy goals: to bring ultra-Orthodox Jewish men, who are routinely granted draft exemptions, into the military, to provide affordable housing and to change the current fragmented multiparty system, which often gives smaller coalition partners outsize strength.

    But Netanyahu only alluded to peacemaking in vague terms, saying coalition talks would focus on "security and diplomatic responsibility." He took no questions from reporters and immediately walked out of the room.

    Netanyahu's comments were clearly aimed at the 49-year-old Lapid, a popular former TV talk-show host who has portrayed himself as an average Israeli and champion of a middle class struggling to make ends meet.

    Though committed to pursuing peace, Lapid's campaign focused heavily on pocketbook issues, raising speculation that Lapid might abandon the peace agenda if he can extract other concessions from Netanyahu.

    In an interview with The Associated Press last week, Lapid criticized Netanyahu's handling of peace efforts, saying he was committed to restarting negotiations and would not serve as a "fig leaf" in a hard-line government.

    Dov Lipman, a lawmaker in Lapid's party, said Yesh Atid was serious about resuming talks with the Palestinians. He said the party's strong performance "clearly says the people of Israel, while focusing on internal issues ... do understand we have to be in negotiations, exploring solutions and have to be trying to get to the two-state solution."

    "We feel that with a sincere approach from the Israeli side, showing we're serious, we can make progress," he said. "We don't feel it's responsible leadership to be in a government which is not trying to move forward in negotiations."

    Talks have ground to a standstill during Netanyahu's past four years in office, in large part because of his continued construction of Jewish settlements in the West Bank and east Jerusalem. The Palestinians claim both areas, captured by Israel in 1967, for a future state and refuse to negotiate while the construction continues.

    Under Israel's election laws, President Shimon Peres must formally charge Netanyahu with the task of forming a new coalition within six weeks.

    If he reaches a power-sharing deal with Lapid, it remains far from clear whether it would be enough to restart talks. While Netanyahu has grudgingly accepted the idea of establishing a Palestinian state, his Likud Party is dominated by hard-liners who oppose Palestinian independence.

    Another potential coalition partner, Naftali Bennett of the pro-settler Jewish Home Party, has called for annexing large parts of the West Bank, the heartland of a future Palestine. Netanyahu will be hard-pressed to balance the competing visions of Bennett and Lapid, and resumption of negotiations would likely spark a revolt within his hard-line base of support. A continued 60/60 deadlock would trigger new elections.

    On the other hand, Lapid's positions, while generous in Israeli terms, still fall short of Palestinian demands for a full withdrawal from all of the West Bank and east Jerusalem. Lapid says east Jerusalem, home to Jewish, Christian and Muslim holy sites, must remain under Israeli control. He has also refused to say whether he accepts a freeze in settlement building, a key Palestinian demand.

    In the West Bank city of Ramallah, Palestinian leaders said Wednesday they were pleasantly surprised by the strong showing by Israel's moderates, but remained doubtful that would soften Netanyahu's policies.

    "You are not going to have a savior suddenly producing instant peace," said Hanan Ashrawi, a senior official with the Palestine Liberation Organization.

    Opinion polls in Israel have indicated a majority favor establishing a Palestinian state alongside Israel. Yet during the election campaign, the need to reach peace was barely mentioned, reflecting widespread doubts, even among the dovish left, that peace is possible.

    The outcome of Tuesday's election shocked Israel, where Netanyahu had been forecast by all opinion polls to be cruising toward re-election at the helm of a hard-line and religious bloc.

    Netanyahu's Likud-Yisrael Beitenu alliance captured just 31 seats, down from 42 in the outgoing parliament. After the shocking setback, Netanyahu found himself answering tough questions about what went wrong.

    Avraham Diskin, a political scientist from Jerusalem's Hebrew University, said Netanyahu's merger with former Foreign Minister Avigdor Lieberman's Yisrael Beiteinu turned out to be a huge error. The union was supposed to create a strong rightist bloc immune from blackmail attempts of smaller parties.

    In practice, Diskin said, it backfired because the parties' constituencies have little in common. Likud appeals to traditional, religious Jews and those of Middle Eastern descent. Lieberman's base are immigrants from the former Soviet Union. Lieberman's indictment on corruption charges during the campaign also hurt the alliance.

    "This was a harsh error. It was the deciding factor," Diskin said.

    Netanyahu made other gaffes. His campaign, including photo ops with young smiling soldiers and stops at Jewish holy sites, appeared to take victory for granted. An attack on Bennett, the settler leader, ended up rallying sympathy for his rival, and the last-minute appointment of a popular Likud Cabinet minister to a senior government post appeared to be a desperate attempt to win over voters.

    In the background, Netanyahu's tense relations with key allies, including the United States, over settlement construction also may have concerned voters. In the weeks before the vote, Netanyahu announced plans to build thousands of new settlement homes. Last week, President Barack Obama was quoted as saying Netanyahu was hurting Israel's own interests with the settlement construction.

    On Wednesday, White House spokesman Jay Carney pushed back on the notion that Obama and Netanyahu need to recalibrate their relationship.

    "No leader has met more often with or spent more time on the phone with President Obama than Prime Minister Netanyahu. That relationship is strong, and it is a relationship that allows for a free and open discussion of ideas and positions," Carney said.

    Sima Kadmon, a veteran political columnist for the Yediot Ahronot daily, said the election results did not represent a a major shift in ideology but rather disappointment in Netanyahu himself.

    "There is no doubt Netanyahu is the tragic figure in these elections. Despite the huge advantage with which he set out ... he ended up as, at best, a weak prime minister whose coalition will depend on the wishes of the big winner in these elections, Yair Lapid."

    The six biggest moments from the season premiere of ?The Ultimate Fighter?

    The latest version of "The Ultimate Fighter" debuted on Tuesday night. For the first time in many, many seasons, I enjoyed the show. Instead of focusing on the animosity between coaches, it brought out the stories of each of the fighters. It was a welcome change. Here were the biggest moments of the two-hour premiere:

    -- The opening montage: From the start, TUF looked different than it had before. The filming style felt more like an "NFL Films" production than previous TUFs. That started with the opening montage, with close-ups of the coaches, Dana White and the fighters talking about why they want to be on the show.

    -- Families fighters and friends being allowed in the gym: Seeing Adam Cella's family celebrate after his win set the tone for the rest of the show. On previous TUFs, fighters would talk about their devotion to their amily and friends, but it was hard to connect with them when all we saw were pictures. Now, as the season goes on, we'll know who each man is fighting for much more richly.

    -- Zak Cummings' knockout of Nik Fekete: It was so quick that I wasn't sure what happened. The replay showed Fekete didn't protect himself when throwing a kick, and that ended his show at making "The Ultimate Fighter."

    -- Sonnen needling Jones over Bubba McDaniel: The "Chael Shtick" didn't come out much on Tuesday night, and that's a good thing. Over the course of the show, it would get old very quickly. But Sonnen's subtle, sarcastic picking at Jones was fun, particularly when it came to Jones' teammate and if Jones would pick him for his team.

    -- "I grew up on food stamps, powdered milk and spam." With the families around, there were more tears than usual for the opening show. No episode was more moving than Kito Andrews saying goodbye to his kids, and talking about why it's so important for him to win. Unfortunately, Andrews lost the decision.

    -- Uriah Hall's decision over Andy Endz: The two put on such a great fight that Sonnen stood and cheered for it. Afterwards, they shared a quiet moment where Hall thanked Endz, and Endz told Hall to win the show. It was one of the moments that reminded fans that most fighters support each other, even when they haven't met before.

    Empire State Building shooting victim sues the NYPD

    NEW YORK (Reuters) - One of nine bystanders shot when two New York police officers fatally shot a homicidal suspect outside the Empire State Building last summer has filed a lawsuit charging that the officers - who fired 16 bullets, killing the suspect - were not adequately trained.

    The tense August 24 showdown, at the height of morning rush hour on one of Manhattan's busiest streets, was captured on video and shows suspect Jeffrey Johnson stepping from behind a concrete flower pot and pointing his gun at two approaching officers, who swiftly returned fire.

    A few minutes earlier, Johnson had shot a former co-worker at close range, killing him. The confrontation with police lasted less than 10 seconds.

    "The NYPD is aware of the deficiencies in its training program," said Stephen B. Epstein, one of the attorneys representing shooting victim Chenin Duclos, 32, a physical therapy student who was struck in the leg, shattering her femur.

    "The reason they haven't made adjustments to the training is that it would cost a substantial amount of money, and they have a budget," Epstein said. "The reality is, they're better off paying off judgments than changing training."

    An NYPD spokesman vigorously defended the department's firearms training program.

    "Actually, it's the best-trained police department in the country," chief NYPD spokesman Paul Browne said on Wednesday, adding that "you won't find anything close to the level of annual training the NYPD goes through" on any other U.S. police force.

    Browne said the department's 35,000 officers receive firing range training twice a year, and complete an annual course in urban military warfare and counter-terrorism at a Bronx police training facility where real-time urban crime scenarios are rehearsed.

    "Any reasonable person that sees that video sees that the officers had no choice but to fire," Browne said.

    The lawsuit contends that the two officers "ran up upon Johnson, pointed at him, called out to him, approached him, and attempted to grab him, causing him to draw his weapon, instead of following at a safe distance, calling for back-up officers, and waiting to confront Johnson at a location away from a crowded street."

    Most of the wounded were hit by shrapnel after police bullets struck concrete flower pots placed in front of the city landmark to protect it from car bombers, and splintered, police officials said. Epstein said it remained unclear if Duclos was hit by shrapnel or a bullet.

    The NYPD's 2011 Firearms Discharge Report said that last year, 35,000 NYPD officers had 23 million encounters with civilians, and discharged a weapon in 92 incidents. Nineteen of the incidents caused injuries and nine caused a death, according to the report.

    A 2008 Rand Corporation study of the NYPD's firearms training program recommended expanding the use of Tasers, sensitizing officers' ears to the sound of gunfire during firearms training, and more frequent overall training.

    At least five of the other eight shooting victims have filed notices of claim with the New York City Comptroller's office, a spokeswoman said, the first step toward filing a lawsuit.

    Oscars: Final Voting Schedule, Ballot Options Announced

    LOS ANGELES (TheWrap.com) - Members of the Academy, grab your ballots!

    The Academy of Motion Picture Arts and Sciences announced the schedule for the final round of votes for this year's Oscars on Tuesday. The Academy also said that it had increased access to paper ballots in order to encourage engagement among Academy members.

    Final balloting for the Oscars begins Friday, February 8 at 8 a.m. PT, and closes February 19 at 5 p.m. PT. Academy members who voted online during the nominations round will also be given the option to use paper ballots in the final round, as will members who registered to vote for the final round online. (Those who registered to vote online have until February 1 to request a paper ballot from the Academy's Membership Department.)

    Voters will also be aided with the use of assisted voting stations in Los Angeles, New York and London, as well as a 24-hour call center.

    This marks the first Oscars that have allowed electronic voting, which has led to a period of adjustment for Academy members. Late last year, the Academy extended the deadline to request paper ballots for the nominations round by two weeks, from November 30 to December14, in order to give voters adequate time to familiarize themselves with the various voting options.

    The outreach effort has apparently worked; the Academy's chief operating officer Ric Robertson said Tuesday that this year's nominations round drew an increased number of votes over previous years.

    "One of our basic goals at the Academy is to increase member engagement. Despite some challenges, more members voted for this year's nominations than they have in the past several years," Robertson said. "We are looking forward to a continuation of that trend in the final voting."

    The Oscars will air live on February 24 from the Dolby Theatre at the Hollywood & Highland Center on ABC. with Seth MacFarlane hosting the program.

    Quadruple DNA helix discovered in human cells

    Sixty years after James Watson and Francis Crick established that DNA forms a double helix, a quadruple-stranded DNA helix has turned up.

    Quadruple helices that intertwine four, rather than two, DNA strands had been made in the laboratory, but were regarded as curiosities as there was no evidence that they existed in nature. Now, they have been identified in a range of human cancer cells.

    The four-stranded packages of DNA, dubbed G-quadruplexes, are formed by the interaction of four guanine bases that together form a square. They appear to be transitory structures, and were most abundant when cells were poised to divide. They appeared in the core of chromosomes and also in telomeres, the caps on the tips of chromosomes that protect them from damage.

    Because cancer cells divide so rapidly, and often have defects in their telomeres, the quadruple helix might be a feature unique to cancer cells. If so, any treatments that target them will not harm healthy cells.

    "I hope our discovery challenges the dogma that we really understand DNA structure because Watson and Crick solved it in 1953," says Shankar Balasubramanian of the University of Cambridge, UK.

    Balasubramanian's team identified the four-stranded structures in cancer cells with the help of an antibody that attaches exclusively to G-quadruplexes. To stop them from unravelling into the ordinary DNA, they exposed the cells to pyridostatin, a molecule that traps quadruple helices wherever they form.

    This enabled the researchers to count how many formed at each stage of cell multiplication. The G-quadruplexes were most abundant in the "S-phase" ? when cells replicate their DNA just prior to dividing.

    "I expect they will also exist in normal cells, but I predict that there will be differences with cancer cells," says Balasubramanian. His hunch is that the G-quadruplexes are triggered into action by chaotic genomic mutations and reorganisations typical of cancerous or precancerous cells.

    "This research further highlights the potential for exploiting these unusual DNA structures to beat cancer, and the next part of this is to figure out how to target them in tumour cells," says Julie Sharp of Cancer Research UK, which funded the research.

    Another important question that Balasubramanian's and other teams will try to answer is whether G-quadruplexes play a role in embryo development, and whether such a role is mistakenly reactivated in cancer cells. "We plan to find out whether the quaduplexes are a natural nuisance, or there by design," he says.
